| I. Background | |
| 激活素 A（Activin A）是轉(zhuǎn)化生長因子-β (TGF-β)超家族的重要成員，是一種多能因子， 參與過敏、自身免疫性疾病、癌癥等免疫失調(diào)疾病的發(fā)生。 激活素 A 通過兩種 I 型受體和兩種 II 型受體發(fā)出信號，當它們與配體結合時，組裝成 最終的受體復合物。I 型受體包括 1A 型激活素受體（或激活素受體樣激酶 2，ALK2）、 1B 型激活素受體（或 ALK4）和 1C 型激活素受體（或 ALK7）。Activin-A 有利于 ALK4 結合，而對 ALK2 和 ALK7 的親和力較低。II 型受體是 Activin 受體 IIA 型 (ActRIIA) 和 Activin 受體 IIB 型 (ActRIIB)，其特征是組成性活性絲氨酸/蘇氨酸激酶活性。一旦激活素 A 與其 II 型受體結合，兩個 I 型受體就會被募集，并被 II 型受體磷酸化，這一事件導致其 激酶活性被激活。大多數(shù) TGF-β超家族成員與 ActRIIB 或 ActRIIA 結合，而只有少數(shù)配體 與 ALK4 高親和力結合。除了典型受體絲氨酸/蘇氨酸激酶- smad 軸外，激活素 A 還被證明 參與 P38、ERK、PI3K、WNT 等途徑。 |
