PDGFRA 基因位于人類(lèi) 4 號(hào)染色體 4q12 位點(diǎn)，編碼血小板衍生生長(zhǎng)因子受體α （PDGFRA），屬于酪氨酸激酶受體家族，正常參與細(xì)胞增殖、存活、遷移及血管生成調(diào) 控；其突變或融合（如 D842V 突變、FIP1L1-PDGFRA 融合基因）可導(dǎo)致受體持續(xù)激活， 驅(qū)動(dòng)多種腫瘤發(fā)生，包括約 5%-15%的胃腸道間質(zhì)瘤（GIST，多見(jiàn)于胃部，對(duì)伊馬替尼敏 感但 D842V 耐藥）、嗜酸粒細(xì)胞增多綜合征（HES，如 FIP1L1-PDGFRA 融合患者伊馬替 尼治療有效率超 90%）、膠質(zhì)瘤及部分特發(fā)性肺纖維化等；檢測(cè)方法涵蓋 PCR、測(cè)序、 FISH、數(shù)字 PCR 及液體活檢（ctDNA 監(jiān)測(cè)）。 |

| II. Description | |
|
BaF3（小鼠原 B 細(xì)胞）的生長(zhǎng)和增殖需要 IL-3 的維持。通過(guò)引入各種表達(dá)激酶的基因， 能作為 BaF3 的驅(qū)動(dòng)基因，讓 BaF3 不再依賴(lài) IL-3 而增殖，進(jìn)而激酶基因成為 BaF3 增殖依 賴(lài)的驅(qū)動(dòng)基因，用于評(píng)估小分子藥物對(duì)激酶的靶向抑制作用，原理及流程見(jiàn)下圖所示。
