| I. Background | |
| IL-1 家族包括 11 種細(xì)胞因子，7 種具有激動劑活性的配體（IL-1α，IL-1β，IL-18，IL- 33，IL-36α，IL-36β，IL-36γ）和 4 種具有拮抗活性的成員[IL-1 受體拮抗劑（IL-1Rα）， IL-36Ra，IL-37，IL-38]。IL-1α，IL-1β，IL-18，IL-33，IL-36α，IL-36β，IL-36γ觸發(fā) MPKK 和 NF-κB 導(dǎo)致炎癥反應(yīng)。 成熟的 IL18 被釋放后，主要通過結(jié)合靶細(xì)胞表面的受體復(fù)合物啟動信號傳導(dǎo)。它首先 結(jié)合 IL-18 受體α鏈（IL-18Rα），隨后招募 IL-18 受體β鏈（IL-18Rβ）形成異源三聚體，這 一過程主要發(fā)生在 T 細(xì)胞、NK 細(xì)胞等免疫細(xì)胞上。該復(fù)合物通過招募接頭蛋白 MyD88， 最終激活 NF-κB 和 MAPK 等關(guān)鍵信號通路，誘導(dǎo)炎癥相關(guān)基因表達(dá)。為防止炎癥反應(yīng)過度，人體內(nèi)存在一個高效的“剎車”系統(tǒng)——IL-18 結(jié)合蛋白（IL-18BP），它能以極高親 和力捕獲游離的 IL-18，阻斷其與受體的結(jié)合，從而精確調(diào)控 IL-18 的生物活性。 |

| II. Description | |
| IL18 Effector Reporter Cell(Adherent)，在由調(diào)控因子調(diào)控并表達(dá)報告基因的重組細(xì)胞上，穩(wěn)定表達(dá)人 IL18Rα和人 IL18Rβ。 IL18 Effector Reporter Cell(Adherent)報告基因藥靶模型很好的模擬了體內(nèi) IL18 的信號轉(zhuǎn)導(dǎo)過程，原理見下圖所示。
